e.g. for 买一辆车至少要一萬块钱。
could one also say
买一辆车以上要一萬块钱。

Not quite. 以上 in this context always comes after the minimum number. E.g. 50岁以上 “Over 50 years old". However, it also means "mentioned previously" used in a piece of writing or speech, in which case it can come first, e.g. 以上是我们公司的基本情况。"Mentioned previously is our company's basic situation."
